Intermedins are a part of the calcitonin/gene-related peptide (CGRP) family. In vitro experimentation has demonstrated that the synthetic 47 amino acid intermedin peptides act via the calcitonin receptor-like receptor/receptor activity-modifying protein (CRLR/RAMP) complexes by increasing the levels of intracellular cAMP. Unlike calcitonin gene-related peptide (CGRP) and adrenomedullin (ADM), intermedin does not exhibit selectivity for any of the three RAMPs when co-expressed with CRLR. Studies on normal and spontaneously hypertensive rats have shown that intermedin treatment leads to a reduction in blood pressure that can be prevented by the CGRP receptor antagonist CGRP (8-37). In addition to its hypotensive effects, synthetic intermedin has also been observed to inhibit gastric emptying and food intake in mice. Thus, intermedin serves as an additional regulator of gastrointestinal and cardiovascular functions, and may also be involved in other bioactivities mediated by the CRLR/RAMP receptor complexes.
